Isolation of phosphate solubilising microorganisms from rhizosphere of chickpea

Abstract:
In the present investigation, forty chickpea rhizosphere soil samples were collected from different locations in Karnataka’s Raichur and Gulbarga districts to isolate phosphate-solubilising microorganisms (PSMs). Totally 80 PSMs, consisting of 40 phosphate solubilising bacteria (PSB) and 40 Phosphate solubilising fungi (PSF), were obtained from the 40 samples used for enumeration of the PSMs population. Among the 40 PSB and 40 PSF strains, ten each efficient PSB and PSF strains were selected based on their solubilisation index in Pikovskaya’s agar media. They were identified up to the generic level. Selected PSMs were grown in vitro for ten days on Pikovskaya’s medium, and the following analyses were carried out i.e. pH change and phosphorus (P) solubilised. P solubilisation index of the twenty best isolates ranged from 1.80-3.17, the drop in pH of the medium ranged from 7 to 3.1, and the amount of TCP solubilised ranged from 84.0 mg/l to 165 mg/l. The study observed that the bacteria have more inorganic insoluble phosphate solubilising ability than fungi.
